Setting up Great Western Trail: Second Edition involves placing the main game board in the center of the table and preparing the various components. Station master tiles are placed along the railway track, neutral building tiles on designated board spaces, and three bags containing hazard and objective tiles near Kansas City. The market board is populated, player boards receive respective player pieces, starting workers, cattle cards, and private building tiles, and all money is organized nearby. Additionally, objective cards are shuffled, and starting capital is distributed based on player count.

The main objective of the game is to guide your herd of cattle from Kansas City along the trail to various railway stations in the east, earning money, upgrading your train, and collecting victory points through strategic deliveries, building placements, and objectives.

On your turn, you move your herder along the trail spaces according to your movement allowance, stopping at neutral or player-owned buildings to perform their associated actions. Actions include hiring workers, building structures, purchasing cattle cards, removing hazards, and advancing your train.

End-game scoring awards victory points for successfully delivered cattle to cities, the furthest reach of your train on the railway track, collected station master tiles, completed objective cards, remaining money, and bonuses from your player board and built structures.

The game concludes when specific end triggers are reached, typically when the market tile deck or worker supply needs to be replenished but cannot, signaling the final rounds for players to complete their current actions.
