Setting up Lorenzo il Magnifico involves placing the main board in the center with its four towers loaded with randomized character, building, venture, and territory cards. Each player takes a personal player board, four colored family member tokens, a neutral beige worker, and initial resources including coins and servants based on turn order drafting. Bonus tiles and leader cards are drafted at the beginning to give players asymmetric starting goals and ongoing abilities.

The main objective of the game is to amass the highest amount of prestige, fame, and victory points by the end of six rounds. Players act as heads of noble families in Renaissance Florence, striving to level up in society through card acquisition, resource management, and fulfilling church expectations.

During each round, players take turns placing one of their family members onto available worker placement spots on the main board. The value of a player's worker is determined by three rolled dice at the start of the round, modified by servants or specific card effects. Actions allow players to collect resources, acquire and activate cards, or take council privileges.

Scoring happens throughout and at the end of the game. Points are accumulated via character card collections, end-game venture cards, military tracking, and leftover resources converted at a 5-to-1 ratio. Additionally, surviving or supporting Vatican reports awards crucial faith-based points.

The game is structured across three distinct periods, with each period containing two rounds for a total of six rounds. After the final round and subsequent Vatican report and end-game scoring, the player with the most total points is crowned the winner.
